Spermine and spermidine are naturally occurring polyamines found in the sperm of animals, including humans. They are fundamental components of sperm cells and are involved in various biological processes, such as cell growth, differentiation, and apoptosis. These compounds have been studied for their potential health benefits, including their role in male fertility and as potential anti-aging agents. They are also used in some skincare products for their moisturizing and protective properties.
Industrially, the primary raw material for spermidine is putrescine (butanediamine), which can be obtained through the acrylonitrile-butanediamine Michael addition-hydrogenation route or microbial fermentation of arginine/ornithine. Spermine is obtained by introducing a propylamine group into spermidine, thus spermidine is both a product and a precursor to spermine. In terms of cost structure, 70% of the cost of the chemical method is spent on reductive hydrogenation, protection-deprotection, and purification. Solvent hydrogenation using Pd/C, Raney nickel, and depreciation of high and low pressure reactors account for the majority of the cost. The fermentation method uses only glucose and inorganic nitrogen as raw materials, but involves multiple enzymatic steps, a product concentration of only 1–2 g L⁻¹, and downstream concentration/decolorization accounts for more than 55%. Currently, the cost per kilogram is still 15–20% higher than the chemical method.
